Healthy snacks and frequent stops. Have lots of water. Don't be afraid to pull over at a stop or hotel for a nap. Sleepiness at the wheel is dangerous.

If you're driving, the only counters really are coffeeine and music. Windows open also helps me stay alert, since the wind noise keeps me from dozing off.
You could also pick up hitchhikers. They generally make for good conversation, and none of the ones I've picked up turned out to be crazy-psycho-murder-rapists.

To be on realistic though,
If you were the driver, I would suggest hydrating, not only with caffeine, but also good amounts of H2O as well, this way you can slow down the process of getting that 'crash' feeling after a lot of caffeine intake.
Also take periodic breaks after 2 hours at least. Trust me, you do not want to be a cause of an accident because you feel asleep during bumper-to-bumper traffic, it's not fun paying $250 insurance even though you were only paying $75 previously...
If you are the passenger, assist the driver by taking over an giving him/her some rest. Also entertain yourself. It's a long-ass ride, you gonna need it. I'd suggest watching the Lord of The Rings Trilogy. You'll either pass time real quick by sitting through every movie, or pass out after a a few hours into it.
Ensure that if you are either one, to rest up early the prior day before the event, as well as have everything prepared prior. I'd hate to see someone bitch about something they forgot to bring before the road-trip.
