It all starts with a yellow rubber duck.
In MGS2, you find it in one of the air pockets [while getting/rescuing Emma]
In Castlevania: Aria of Sorrow there is a room with Succubus and Lilith enemies

So what I'm saying here is, there was a good chance that Konami was hinting at a Kojima Castlevania years before it was even announced [This game was Lords of Shadow].
ITT: I can't look at Konami the same way again.

I doubt it, there's a rubber duck in Castlevania: Order of Ecclesia as well, at Somnus Reef, and one in Harmony of Despair. There's probably more of them in other games, but I haven't played that much Castlevania.
As for there being one in MGS2, idk, check some more Konami games and see what comes out of it, it might be a running gag in Konami games in general.
