I see pokemon D/P is popular and is a positive game for pokemon fans so if your a fan you should get it, anything else depends on what genres you like.

Buy a flash card, then you can test out as much as you want. I got my R4 (1 gb micro sd card + actual ds cartridge it goes into) for $80 Canadian.
So, I just download whatever I feel like it whenever I feel like it.
Pokemon D/P (yay, 999 hours =D)
FF III (I beat it, but it was boring for me T_T; guess I'm not the FF type of guy)
Elite Beat Agents (beat it to the max =D)
Osu! Tatakae! Ouendan (J-version, came before EBA, beat it to the max as well =D)
Moero! Nekketsu Rhythm Damashii Osu! Tatakae! Ouendan 2 (J-version 2, came after EBA, beat it to the max as well =D)
Tetris DS (the only game out there to ever compete with my addiciton towards Pokemon)
When I'm at school, linking with friends or even random people (happens alot in a boring class), there are always two games that's a standard: TETRIS and MARIO KART.
I beat a lot more games and enjoyed them as well, but these are just some I can pick off at the top of my head.

Phoenix Wright: Ace Attorney, Justice for all, and (in september) Trials and Tribulations should all be at the top of the list. Awesome games.
Trauma Centre is a surgical sim. It starts off normal and then eventually you have to fight weird alien looking viruses so it sort of strays from reality.
Castlevania Portrait of Ruin. It's Castlevania. Get it.
New Super Mario Bros. It's a Mario game so there isn't anything to explain.
Advance Wars Dual Strike. It's out of print so you may not be able to find it without an eBay account but it's a great game.
Puzzle Quest: Challenge of the Warlords. It's bejeweled with RPG elements. It's an odd game but it's $30 Canadian so you can get it cheap.

Someone's been watching too much Harvey Birdman: Attourney at Law. Back on topic, if you wouldn't mind imports, I suggest Jump Super Stars, but more importantly, I suggest Jump Ultimate Stars above all. All your favorite Shonen Jump characters packed into one game that plays like Super Smash Brothers...except...better. Also, Jump Ultimate Stars has Wifi play, so you can attempt to kick Japanese player's ass... Mind you, attempt... You may or may not succeed. If you decide to get them, there are translation FAQs on Gamefaqs for them.

Definitely get Jump Super Stars, its an awesome fighting game and even though its an import, some Best Buys carry it (got mine in one in NY).
Other then that, RPG-wise definitely FF and Luminous Arc (just finished it and it was great). Finally, the classics, including Super Mario, Mario Kart, and Metroid.
