Have you tried deleting the original 160.doc and 2.doc files off your desktop first? Those documents are prefixed with ~ and set as hidden files because they are backups for currently opened Word documents.
Desktop.ini is a Windows system file that stores information about customized folders. If you have customized the display of a folder in any way, such as changing its icon or manner of display, Windows will save those settings in a desktop.ini file. Yes, your Desktop is in fact a folder.
I would suggest you stop working from your Desktop and move your documents to the system's "My Documents" folder, it is there for a reason.
Or you can disable the "View Hidden Files and Folders" option.

Its actually not the view hidden files and folders option as desktop.ini belongs to the protected operating system files.
Just open up an explorer window like you have computer opened in the screenshot and open folder options and in the view tab make sure "Hide protected operating system files" is checked.

Desktop.ini is not a protected system operating file, as it is in just about every folder with any thumbnail images. It's commonly found within picture folders or music folders that display album artwork.

Desktop.ini only shows up for me when I uncheck hide protected operating system files. I have hidden files set to be seen as well.
This guide explains in more detail how to hide it.

What operating system are you using?

To OP, it would be wise if you run a full AV scan on our computer.
That file shouldn't be appear if you didn't change any settings on your Folder Option.

I believe Nachbar is correct about desktop.ini being categorized by Windows as a "protected system file". Also, it seems to have vestigial purpose, as deleting it doesn't appear to affect your system negatively.
