And I've reached 21 plays on Auto-Play but whenever I press the button there the quest says I've still got to finish it.
Know why?
I think it's the same case with the ONE hour quest never ending even if you play all night. But I'd recommend you to play Auto-battle 2 or 3 more times in case you miscalculated.
If it's really bugged or something, you'll have to make a thread on the support/report forums of SGO site about it.

I think your problem is rather obvious. You need to participate in auto-battle 20 times, not to matches 20 times in it. And now I have proof too:

In order to anticipate new arrivals, the leagues will only run for... I'll get to that if people show interest.
But no, there won't be a DP limit. Unlike the tournaments, this league will reward quality.
I'll think and rules and shit if it picks up interest. Might allow more engaging competition over a longer period than a tournament.

hmm... well for the most part I think I am getting the hang of battles, I feel like I need to farm that beginner dungeon a few times though, and upgrade a few of my cards. But other than that I think I'll be alright.
