To play well in Guitar hero U need 3 Things..
Mind, Body & Soul....
For mind...
*1 To play well Ur mind is important. When playin Concentrate On the notes Ignore the background.
*2 Get a good Tv. U don't want to play on a Lousy Tv That jumps every 10 sec right??
*3 Get a comfortable place to sit of course!! u don't wanna sit on something Thats stickin in your ass!!!
Now for the Body...
1) Warm ur fingers up do some stretching on them So u can Run them well on the guitar.
2) Drink plenty of water and Check ur time don't play too long And of course Remember to do your "business"...
3) Room's temperature must be just nice don't Freeze your self in the room!!!
Lastly on Soul...
#1 Put Ur Tv volume to the MAX Feel The Music!!!! Just remember not to disturb your neighbor...
#2 Do Not Do Stunts with your guitar Unless u are using the controller...

Cracking your fingers to loosen them helps. Well when playing real instruments to be exact, not sure about using a peripheral.

Cracking your knuckles isn't relevant.
Being able to read the game's notation very quickly is important. There's not time to process every note, so you have to be able to process patterns and blocks of notes. You also have to have mapped preferred fingering patterns for certain blocks so that you can access them instantly.
Rhythm and keeping the beat of each song are also important.
If you are making mistakes on a fast section, you are probably rushing.

My friend plays the acoustic guitar and cracks his knuckles on a daily basis and so far he haven't had any problems. Along with my other friend who plays the piano, commented that it actually loosens his fingers and he can play it with ease. I have yet to believe that cracking your fingers causes any major problems for a musician other than loosening your grip. Being a baseball pitcher who cracks their knuckles is another story.

Hmm, I can add more to this list of advice :)
If you're having trouble going from one finger combination to another, do it v e r y slowly about 10 or 12 times. Rest your fingers for 20 seconds or so, and do it again.. SLOWLY. Then speed it up, try it out faster a few times. Fingers still don't want to do it? Go play something else, then come back and do it sloooowly again. It's all about muscle memory, if your fingers remember where to go without you thinking about it, your mind can be on other things... like the timing and beat.
