I Need Help.

"She calls me big brother for reasons I can't understand"
You should have taken that as a hint dude. You fell into the brother/friend zone that all guys do not want to fall into when they are chasing the women they like/love.
Move on, like many said.

you're a nice guy...I'm sure you can find a nice girl for yourself...

Grow a pair and suck it up dude.
She obviously don't think of you in a romantic way and most-likely never will. Neither does it sound like she even wants you around her at all.
So leave her alone and just let it be. Don't be a creep.

Spoiler:
Talk to her about it. Ask her the reason for her words and stuff.
Whatever serious happens, APOLOGIZE.
You don't need any reason, just apologize.

I'm sorry. You just have to move on.
It's horribly, well, horrible. On top of that, she's even explicitly said, "don't talk to me anymore"
When a girl says no, even if she doesn't even utter the word. That's that. It's final, finite.
It's over, dude.
Just man up, move on, and just keep going strong.
If she avoids you now, that's also a bad sign. When it's a guy that a girl just can't stand, she avoids him at all costs.
I'm not sure whether or not it's because you've reaad a bit too much manga and watched a bit tooo much anime, but feelings don't really develop that way. Even if you are childhood friends, which is a plus, but really, all that means is that you had to make your move much more earlier than do it now. Being childhood friends just means you get friend-zoned even easier. It's a sad affair, I know, but like everyone's said here, well mostly everyone, get a grip on yourself, move on with life.
Don't do cheesy things like, "Oh, I just want to stay friends with her so I can check up on her every now and then."
Just don't,
1. You'll forever become that creep she's "friends" with.
2. It'll be even more annoying for her
3. You'll end up feeling even more drained, emotionally invested, etc.
See, if you're coming to other people for advice, then you reek of desperation. I'm not saying that a desperate man should be patronized and ridiculed when all he's done is ask for advice, but a desperate man will always be a very unappealing man when it comes to the women he's around and when it comes to love and feelings.
You're all on your own, because it's all about your self-image and your gut feelings. Doing what you think is right, because you're the only one who knows what to do. You're you, of course!
Honestly though, I'll reiterate this message many times.
"Move on with life, you'll get back on your feet"
K?
